This amendment to Form 8-Kis being filed to amend and supplement a Form 8-Kfiled by C4 Therapeutics, Inc. (the "Company") on January 10, 2022 (the "Form 8-K")solely for the purpose of correcting the hyperlink to Exhibit 99.1 contained in Item 9.01 of the Form 8-K. Other than the foregoing, there are no changes to the disclosures provided in the Form 8-K.

By leveraging its proprietary Target ORiented ProtEin Degrader Optimizer (TORPEDO) platform, to design and optimize small molecule protein degraders that are active against their desired targets by harnessing the bodyâs natural process for destroying unwanted proteins. Its TORPEDO platform has the capability to design two types of protein degraders and both degrader approaches are reflected in its clinical pipeline. Its product candidate, CFT7455, is an orally bioavailable MonoDAC degrader of protein targets called IKZF1 and IKZF3, which are in clinical development for multiple myeloma and non-Hodgkin lymphoma. Its CFT1946 is an orally bioavailable BiDAC degrader designed to be potent and selective against BRAF V600X mutant proteins. It is also developing CFT8919.
